The SARA consortium addresses the characterisation of ADP receptor signaling in human platelets by combining molecular biology, (bio)medicine, quantitative proteomics and bioinformatics. In an iterative strategy changes in phosphorylation upon specific stimulation/inhibition of platelets from patient material will be absolutely quantified by mass spectrometry and SH2-domain profiling. Generated data will be interpreted by state-of-the-art bioinformatics to allow for accurate modeling of the involved signaling pathways based on time-resolved quantitative data. The resulting model will vastly extend our understanding of platelet activation and additionally drug action thereon. Support of developed hypotheses by functional studies on platelets and clinical data will lead to novel targets for diagnostics and therapy of platelet-derived diseases.
